Animal Cell Culture Flashcards
Animal cell culture
When cells from an intact organism are derived and isolated and maintained and grown in vitro aseptically containing a mixture of nutrients and growth factor.
Primary cell culture
A primary cell culture is when the cells are derived directly from the organism
What is the lifespan of the primary cell culture?
Limited
Why does the primary cell culture have a limited lifespan?
Because as they progressively divide the telomere shortens each time
Name two types of cell culture
Attachment culture and suspension culture
What is attachment culture?
When the cells need an attachment or anchorage to survive and grow
Give example of attachment culture
Kidney cells
What is suspension culture?
When the cells do not need any surface to grow
Give example of suspension culture
Lymphocytes
What is a cell line?
When a primary cell culture is subcultured
Two types of cell line.
Finite and infinite cell lines
What is finite cell line?
The cell line that eventually undergoes senescence
What is infinite cell line?
The cell line that can be subcultured indefinitely
Example of finite and infinite cell lines
Finite - mammalian cells
Infinite - cancer cells or transformed cells
What properties do finite cell line shows?
Contact inhibition, density limitation and anchorage dependence
Multilayer culture formed in?
Infine cell line
First cell line derived from?
Mouse cells
Lag phase
Period of zero growth
Cells get adapted
Undergoes trypsinization, secrete matrix aid for attachment
Log or exponential growth phase
When the cells start doubling on the basis of their growth rate and seeding density
Plateau or stationary phase
When there’s no more nutrients left and all the space has been occupied
A typical animal cell culture media contains?
Amino acids, organic and inorganic acids, hormone, growth factor, vitamins, glucose and serum
Two types of cell culture
Artificial and natural
Natural media
Consist of biologically natural fluid
Disadvantage of natural media
Poor reproducibility
Artificial media
Components are prepared artificially by adding particular solvents (nutrients)
Types of artificial media
Serum containing media
Serum free media
Chemically defined media
Protein free media
Name the most common type of serum
Fetal bovine
Function of serum
Stimulates cell growth and cell activities
Enhance cell attachment
Provide transport proteins carrying hormones
Serum free media is used for?
To culture a specific type of cell line
Chemically defined media
Media contains ultra pure inorganic and organic, pure protein. It is contamination free
Protein free media used for?
Doesn’t have proteins
For superior cell growth and protein expression
