Nitro-Blue Tetrazolium Test
Measures the oxidative burst and diagnose CGD
As respiratory burst occurs, the hydrogen ion reacts with nitro-blue to form insoluble blue crystals
Cytochrome Reduction
When NADPH oxidase is activated in respiratory bust, it produces a reactive oxygen species that oxidize DHR 123 into rhodamine 123 (green)
80 to 100% fluorescent neutrophils are normal, declines with age
Serum Iron
Measures total iron in the serum
Does not correlate with total body iron stores
50 to 10 ug/dL
Transferrin
Measures the amount of transferrin in the serum (transports iron)
Not often used
TIBC
Total iron binding capacity
Measures the total amount of iron that could be bound by protein per dL of serum
150 to 350 ug/dL
% Saturation
Percent of transferrin that is carrying iron
(serum iron/TIBC) x 100
Male 20 to 45%
Female 15 to 45%
Ferritin
Measured as indicator of iron stores
Male 20 to 300 ng/mL
Female 12 to 200 ng/mL
sTfR
Transferrin recpetor in the serum
Correlates with the amount of transferrin receptor being expressed on cells throughout the body
Greater need for iron leads to high levels of transferrin receptor
1.15 to 2.75 mg/L
Lead
Measures lead levels to detect lead poisoning
Homocysteine
Used to diagnose folic acid/cobalamin deficiency
Methylmalonic Acid
High means you have a vitamin B12 deficiency
Schilling Test
Distinguish pernicious anemia from other causes of B12 deficient anemia
Haptoglobin
Measures level of haptoglobin in the blood
Attaches to free hemoglobin
Levels can detect when RBC are damaged/destroyed
DAT
Direct antiglobulin test
Test for the presence of auto antibodies in the blood
Osmotic Fragility
Used to measure erythrocyte resistance to hemolysis while being exposed to varying levels of dilution of a saline solution
