what is the difference between left and rightshift
left shift is an increase in wbc immature cells (more bands and blasts) while right shift is the increase in number of mature cells in circulation
what ids the clinical significance of left shift
it is observed in most infextions and in some cancers
what is the clinical significance of right shift
it is seen in liver diseases, megaloblastic anaemias, haemolitic disorders and allerges
explain the preparation of thich smear
- obtain a drop of blood from EDTA bottle of from finger plick
- place a drop of blood on the slide and spread it using a pasture pipete making a smear
- the slide is left to dry
explain how staining is done
stain the slide into fields stain A (methylene blue) it stains the nucleus to purple
wash the slide for 3 seconds, agitate gently
stain the slide into fields stain B (Eosin) that stains cytoplasm to pink/red for 3 seconds and wash gently withtap water
drai the slide and let it dry
explain th examination of a thick smear
- switch on the microscope and place the stained blood film on microscope satage
- place a drop of immersion oil on the thick film
- using x100 lens(oil immersion) focus on the blood firm
- count up to 100 WBC and take note of white cell morphology
- report the findings in percentage or absolute number
