What is megaloblastic anaemia
Is a disorder characterized by defective nuclear maturation caused by impared DNA synthesis
What causes megaloblastic anaemia
- Vitamin B12 and folate deficiency
Function of vitamin b12 and folate
They are involved in dna synthesis
2 important roles played by vitamin b12
- Acts as co enzyme for mythyltrasferase
- Important in conversion of methylmalonyl
Vitamin B12 sources
- Produced by micro-organisms and fungi
- Present in foods of animal origin
What 2 compounds makes up vitamin b12
- Cobalamin
- Water soluble
What 2 proteins are involved in the transport of vit B12
- Intrinsic factors
- Transcbalamins 1,2
Explain classes of vitamin B12 deficiency
- Due to intrinsic factors

Where do we obtain folate
- Green vegetables
- Absorption through the upper small intestine
What are the causes of folate deficiency
- Nutritional
- Excessive utilisation
- Malabsorption
What are the lab findings of megaloblastic anaemia
In FBC
1. MCV>95 fl
2. White cell count, platelets and retic count is reduced
In blood firm
Oval macrocytes
Serum vitamin b12 is reduced
What the other causes of macrocytosis
- Alcohol
- Liver diseases
- Drugs
- Pregnancy
- Smoking
