W4 microscopy Flashcards
light source
radiant field diaphragm adjusts the intensity of brightness and it is located above the light source controls diameter
condenser assembly
located above light source and just under slide.
the condenser acts to focus light onto the slide on the stage. an adjustable aperture diaphragm controls the angle of the cone of light reaching the specimen the alignment is what the kohler illumination is
stage
where slide placed for viewing.
slide controls
move stage in a horizontal plane
coarse focus knob
moves stage up and down
nose piece
holds objectives
objectives
different settings 10x 40x and 100x magnify the specimen
parfocal
nosepiece and objective assembly that allows different objectives to focus at the same place. you won’t need to move coarse knob only fine focus knob
body tube
enclosed tube that physically separates the objective from optical lens
oculars
located in adjustable binoculars. provide 10x magnification
primary magnification and ocular lens
this is done by the objective lens and the object is seen as real, magnified and flipped. and this image coming from the objective lens is what the ocular lens magnifies which creates a virtual, magnified and upright image
total magnification of microscope
multiply magnification of two lenses
refraction
light bends as it passes through mediums of differing densities the refractive index measures the density of a medium
immersion oil
immersion oil used to prevent loss of light due to refraction and ensure adequate light is collected y smaller diameter lens
resolving power
ability of a lens to discern fine detail and produce a clear image of the specimen
