SOP volumetric pipette
- hold in dominant hand and insert tip below surface of liquid
- hold pipette bulb in other hand, expelling air and put on top of pipette and release pressure to draw liquid past calibration mark
- maintain bulb pressure and lift off and place index finger on top of pipette firmly
- wipe tip of pipette with tissue and discard
- touch tip to see of beak, hold beaker at a light angle and let water flow out– don’t re-emerse tip
- make sure calibration mark is at eye level and allow liquid to drain until meniscus is on calibration * do not wipe tip again.
- touch the tip to the side of the receiving make sure you are holding vessel at angle and keep pipette vertical
- remove finger completely and will pipette to drain freely
SOP Mohr pipette
meassure fractions of a total volume
1. insert tip below liquid surface
2. use a bulb to draw the liquid above calibration mark which would be zero line
3. wipe tip with tissue
4.touch tip to side of beaker with beaker at angle and pipette vertical and lower meniscus to calibration line
5. touch tip to side of receiving vessel
6. raise the calibration mark of the amount delivered to eye level. relax some of the pressure from index finger to allow the liquid to flow to the measured calibration mark and
7/ repeat step 6 for how many receiving vessels you have.
8.stop at lowest calibration line
9. excess reagent disposed of in waste beaker
SOP pasteur pipette
- squeeze bulb of pipette
- insert tip into fluid
- slowly release bulb to draw liquid into pipette. As the bulb part is part of the pipette it is not a concern of the liquids get into the bulb portion of the plastic one.
- slowly squeeze the bulb again to expel liquid from pipette into receiving vessel single drops can be used
MLA pipette
- firmly attach disposable pipette tip
- fully depress the plunger before entering the sample
- immerse the tip into the sample far enough not to draw air but do not go above the pipette tip
- smoothly release the plunger to draw sample into tip
5/ withdraw pipette by sliding the tip up along the wall of the container be careful not to flick the pipette tip on the top edge of the container because that will create aerosols - quickly check for air bubbes
- place tip close to liquid and against the side of the wall of the receiving vessel.
- smoothly depress the plunger
- with plunger depressed withdraw pipette sliding it up along the wall of the vessel
- release the plunger
- remove the used tip with an absorbent pad
- use a new pipette tip each time
SOP eppendorf/ hamilton pipettes
- firmly attache pipette tip
- keeping pipette vertical press plunger to the first stop
- immerse the tip into the sample
- slowly allow the button to glide back and slide the tip out along the inside of the vessel
- quickly check the tip for bubbles
- hold the tip close to the liquid and against the inside wall of the receiving vessel.
- press the button down to the first stop and wait a couple seconds then press the button to the second stop
- with the plunger still depressed withdraw the pipette by sliding it up along the wall of the vessel
- the tip can be ejected by pressing the button to the final third stop or by depressing a button on the side depending on the model
- a new pipette tip will be used each time
