what is a contaminant?
a polluting or poisonous substance that makes something impure
Aseptic technique?
used in microbiology to exclude contaminants
the purpose is to prevent contaminiation
sterile / sterilization
all culture medias which are used to grow microorganisms are sterilized in which they are rendered free of all life
Broth culture?
liquid mixture that provides large numbers of bacteria in a small space and are easily transported
agar slants
are test tubes containing solid culture media that were left at an angle while the agar solidified
agar deep
agar is allowed to solidify in the bottom of a test tube
what is the importance of agar deep?
they are used to grow bacteria that require less oxygen than is present on the surface of the medium.
Aseptic inoculation technique from broth to slant?
Sterilize Loop with other hand take cap off and hold tube at an angle then sterilize the top of the tube get a loopful of culture heat the mouth of the tube, replace cap
Deep and broth
sterilize needle with other hand take cap off and hold tube at an angle
then sterilize the top of the tube
stab agar deep with needle
heat the mouth of the tube, replace cap
slant to slant
inoculate slant by streaking the loop back and forth across the surface of the agar
What is the purpose of flaming a loop and why must it be cooled before collecting organisms?
to sterlize and kill an bacteria and avoid contamination
you must wait for it to be cooled so that it wont kill off what you are trying to inoculate.
