Topic 21 Flashcards
Equation to calculate the Resolving power
1/d = sin u/λ
u = angle of the outermost beam
What are the Rayleigh’s Criterion?
The point where resoluition is obtained can’t be determined but seems to be where the central maximum of one airy disk falls onto the first minima of the other air disk.
Distance between these two points in the resolution limit/resolving power.
Give the Abbe’s Criterion
At least 2 different beams should enter the objective to see any detail in a microscope
Equation to calculate the Numerical aperture of a microscope (Abbe’s Law)
1/d = (n sin u)/λ
n sin u = Numerical aperture of the objective
Describe the immersion microscope
Drop of oil to increase numerical aperture between the onjective and the object.
Describe the UV Microscope
Small wavelengths to improve resolving power.
Cannot be seen with the eye
