What is the mechanism of action?

A

They inhibit the Na+/Cl− co-transporter in the distal convoluted tubule of the nephron.

Preventing sodium reabsorption and its osmotically associated water

What are the side effects of these drugs?

A

Hyponatraemia
Hypokalaemia- causing cardiac arrhythmias
Impotence in men
May increase plasma concentrations of glucose, LDL and triglycerides

A

Gout- as they reduce uric acid excretion precipitating an acute attack
