Reticulocyte Count Flashcards
After the orthochromatic normoblast loses its nucleus, a small amount of RNA remains in the red blood cell and the cell is known as _____.
Reticulocytes
After the _____ loses its nucleus, a small amount of RNA remains in the red blood cell and the cell is known as reticulocytes.
Orthochromatic Normoblast
To detect the presence of RNA, the red blood cells must be stained while they are living and this process is called _____.
Supravital Staining
Materials needed for Reticulocyte Count:
Slides
Anticoagulated Blood
Supravital Stain
Pipette
Microscope
Cedarwood Oil
Types of Supravital Stain to color the Reticulocytes:
Brilliant Cresyl Blue
New Methylene Blue
List down the step-by-step procedure for Reticulocyte Count:
- Place three drops of reticulocytes stain in a small test tube.
- Add three drops of well mixed whole blood to the tube containing the stain.
- Mix the tube and allow to stand at room temperature or incubate at 37C for 15 minutes.
- After 15 minutes, mix the contents of the tube well and prepare smears and allow it to dry.
- Examine the smear under the oil immersion objective.
- Count the total number of reticulocytes found per 1,000 RBCs.
How many drops of Reticulocyte Stain is added on the test tube? (According to the test done in actual laboratory setting)
5 drops of Reticulocyte Stain
How many drops of well-mixed whole blood is added into the tube with reticulocyte stain? (According to the test done in actual laboratory setting)
5 drops of well-mixed blood
The reticulocyte stain with well-mixed blood in a tube is mixed together and allowed to stand at what temperature or it can be incubated for how many degrees and minutes?
Allowed to stand at Room Temperature
Incubated at 37 degree Celsius for 15 minutes
Formula for Reticulocyte Count:
(Number of Reticulocyte Count/1,000) x 100
Reference Range for Reticulocyte: (Adult)
0.5% - 1.5%
Reference Range for Reticulocyte: (Newborn)
2.0% - 6.0%
