Hematocrit Determination Micro Method Flashcards
Materials needed for Micro Hematocrit Testing:
Capillary Tube
Sealing Clay
Microhematocrit Reader
Microhematocrit Centrifuge
What color of Capillary Tube is used for Microhematocrit Test?
Capillary Tube with Blue Band
Why a capillary tube with red band is not recommended for Microhematocrit Test?
This is because the blood is already stored in an anticoagulated tube (EDTA) and when it is placed in a capillary tube with red band, the anticoagulant would be doubled resulting in false results.
When placing the capillary tube with blood sealed with clay and wax, what is the placement in the microhematocrit centrifuge?
The clay ends must facing the outside. The clay must be touching the rubber.
How much blood must be filled in the capillary tube for Microhematocrit Test?
3/4 full
How do we seal the capillary tube used for Microhematocrit Test?
We seal the part of the capillary tube with color band by holding the tube horizontally, seal with clay, and rotate slightly preferrably a 90 degree angle.
Speed and time for centrifugation for Microhematocrit Test?
5 minutes at full speed of 12,000 rpm
Sources of error in Microhematocrit Test:
- Incomplete sealing of the microhematocrit tubes - falsely low results
- Inadequate centrifugation
- Time and speed of centrifugation
- Overanticoagulated
