What are artifacts?
elements that are seem in the microscope but not present in the actual sample
Light microscopy uses what kind of microscope?
compound light microscope (uses an objective lens that is the closest to the specimen and an ocular lens that is located in the eye piece)
How do you calculate magnification?
multiplying the ocular lens magnification and the objective lens magnification together.
What is the general principle of resolution?
The shorter the wavelength of light the better the resolution will be.
Resolution
The ability to distinguish fine detail and structure (how far two points are from one another) light must pass between the objects in order to be seen as seperate from one another
Electron Microscopes
Use beams of electrons which have waves much shorter than those of light therefore they have a greater resolution and magnification
When do we use scanning electron microscopy vs transmission electron microscopy?
SEM microscopes can be used to see the cell membrane and must use a heavy metal stain whereas TEM are used to see the organelles and interior cell structure (cell wall must first be cut and then sections must be stained)
