Purpose of Microscope
create magnified image of objects to small to be seen with the eye only
What does Brightfield microscope use to view specimens?
Light and Glass
Purpose of Oil Immersion
prevents refraction; reduces light scatter and increases resolution; directs more light to objective lens
What objective is immersion oil used with?
Can only be used with 100x objective

Parfocal means
when one objective lens is in focus, the other objective lens will also be in focus

How do you create a wet mount?
Using pipette, drop liquid in center over slide, use cover slip and avoid air bubbles
Special Type of Wet Mount technique where a depression slide is used, and drop of medium is covered by a cover slip

What are the purposes of stains?
Stains are used to see cell structures by enhancing contrast
Directional/ purposeful motility is a
positive test/ true motility, as most motile organisms change position w/ respect to one another (compared to Brownian movement)
