MB - Enzyme Strategies and Proteases Flashcards
1
Q
What are the 2 types of enzyme kinetic mechanisms?
A
- Sequential
- Ping-pong
2
Q
What is the difference between the 2 kinetics mechanisms?
A
- Sequential involves the binding and catalysis of multiple substrates at the same time
- Ping-pong involves the binding and catalysis of one substrate at a time
3
Q
What are the 4 general enzyme strategies?
A
- Position the reactants correctly for interaction
- Distort the reactants making them less stable
- Stabiliste the transition state
- Change the environment to favour the reaction
4
Q
What happens when theres no enzyme present?
A
Substrate molecules arrive from all angles and so are unlikely to bind
5
Q
What does the addition of an enzyme help with?
A
- Substrate molecules can only bind from one direction and hence are more likely to attatch
- Enzymes also destabilise the substrate and stabilise the transition state
6
Q
What are the 3 Specific Chemical Strategies?
A
- Covalent catalysis
- Acid/base catalysis
- Metal ion catalysis
7
Q
What is proteolysis?
A
Where peptide bonds are cleaved by boiling in 6M HCl acid for 24 hours
8
Q
What are serine proteases?
A
Enzymes that cleave peptide bonds, in which serine acts as the nucleophilic amino acid
