The loading dose:
The amount of drug given initially to obtain the desired plasma concentration as soon as possible, a loading dose is usually expressed in mg/kg
The maintenance dose rate:
The rate at which drug needs to be administered to maintain the desired plasma concentration, a maintenance dose rate is usually expressed in mg/kg/min
Loading Dose and Maintenance Dose Rate are both related to desired
plasma concentration and bioavailability
to work out dose interval we need to know
- elimination half-life
- therapeutic window
- compliance
the time it takes for the plasma concentration to halve on account of the drug being eliminated from the body
Depends on two things:
- clearance
- vod
Loading dose uniquely related to
volume of distribution
Maintenance dose rate uniquely related to
drug clearance
