Lecture 15. Antibody Genetics Flashcards
What is an antibody ?
Secreted molecules of adaptive immune system
What are the antibody effector functions ?
- Binding
- Phagocytosis and ADCC
- Complement activation
What does Fab stand for ?
Fragment antigen binding
What does Fc stand for ?
Fragment crystalisable
What is the process of antibody dependent cell cytoxicity ?
- Antibodies bind antigens on the surface of target cells
- NK cell CD16 Fc receptors recognise cell bound antibodies
- Cross linking of CD16 triggers degranulation into a lytic synapse
- Tumour cells die by apoptosis
What type of affinity does an antibody that binds tightly have ?
High affinity
What type of affinity does an antibody that binds loosely have ?
Low affinity
What is recombination signal sequences recognised by ?
RAG (recombination activation gene) products
What cells of B-cell differentiation have RAG ?
- Pro-B
- Pre-B
What cells of B-cell differentiation do not have RAG ?
- B cell
- Plasma cell
What heavy chains do B-cell differentiation cells have ?
- Pro-B have DJ
2.Pre-B have VDJ - B-cell have VDJ
- Plasma cell have VDJ
What cells have light chains (VJ) ?
- B-cell
- Plasma cell
What are hypervariable regions ?
- Interact with antigen
- Sites of affinity maturation
What is another word for affinity maturation and when does it happen ?
- Somatic hypermutation
- Class switching
How is antibody diversity generated ?
- Heavy and light chains
- Combinatorial diversity- gene segments
- Junctional diversity
- Diversity gene segments
- Somatic hypermutation
