Warfarin MOA
inhibits epoxide reductase (reduces Vit K to activate Factors II, VII, IX, X & Proteins S & C) (inhibits pro-coagulation factors)
Heparin MOA
activates antithrombin (which inhibits activated forms of procoagulation factors: Factor IIa (thrombin), VIIa, IXa, Xa, XIa, XIIa) (activates anti-coagulation pathway)
tPA
thrombolytic catalyses reaction of plasminogen to plasmin which cleaves the fibrin mesh into fibrin degradation pathways.
Aspirin
Inhibits cycloxygenase (enzyme that transforms arachadonic acid to thromboxane A2) decreases aggregation of platelets
Clopidogrel & Ticlopidine
inhibit ADP-receptor-induced expression of GpIIb/IIIa (decrease GpIIb/IIIa expression on platelet surface) decreases aggregation of platelets/ linking
Abciximab
inhibits GpIIb/IIIa directly cannot bind fibrinogen and aggregate platelets
Hereditary Spherocytosis
Splenectomy is curative (post-splenectomy will have howell-jolly bodies)
Paroxysmal Nocturnal Hemoglobinuria
Eculizumab
Sickle Cell Disease
Hydroxyurea (increases HbF) bone marrow transplant
Lead poisoning
Dimercaprol EDTA
Acute intermittent porphyria
