What mutations give rise to primary myelofibrosis?
JAK2 or MPL mutations
What are the characteristics of primary myelofibrosis?
It occurs in individuals over the age of 60, usually with anaemia and splenomegaly. Blasts are observed in the peripheral blood, along with normocytic anaemia. Median survival is 3–5 years, due to increased risk of infection, as well as transformation to AML in around a fifth of cases.
What are the causes of microcytic anaemia? (TAILS)
Thalassaemia, anaemia of chronic disease, iron deficiency, lead poisoning, sideroblastic
What are the causes of macrocytic anaemia? (FATRBC)
Fetus (pregnancy), alcohol, thyroid (hypothyroidism), reticulocytosis, B12 and folate deficiency, cirrhosis and chronic liver disease
