Genetic Technology: Gel Electropheresis & Polymerase Chain Reaction Flashcards
1
Q
DNA Fingerprinting
A
the use of technology to identify an unknown source of DNA
2
Q
What are the basics of DNA fingerprinting?
A
- Isolate the DNA
- Make copies (if needed) - via PCR
- Cut DNA up using restrictive enzymes
- Compare the fragments form step #3 to known samples - via Gel Electropheresis
3
Q
What are the first three steps in PCR?
A
- Heat DNA sample to unwind it
- Design and add primers to base pair with DNA strand
- Add DNA polymerase and nucleotides
4
Q
After step three when the DNA polymerase and DNA nucleotides are added, what are the last steps?
A
- Cool the mixture of DNA, primers, nucleotides
5. Repeat the process to make desired number of DNA copies.
5
Q
Gel Electropheresis
A
separates macromolecules (nucleic acids and proteins) on the basis of size and electrical chage
6
Q
What are the applications of gel electrophoresis?
A
- crime scene analysis
* DNA sequencing
7
Q
What are the three steps in electrophoresis?
A
- Treat a DNA sequence with a restrction enzyme
- Load DNA into a gel and turn on electric field (running negative to positive)
- Gels are stained and banding patters are observed.
