What is the advantage of examining fresh tissue?
Protoplasmic activities can be observed / viewed
What are the exs of protoplasmic activities?
1) Motion
2) Mitosis

What is the process of teasing or dissociation?
Tissue sx is immersed in a watch glass w/ an isotonic saline solution (dissected or separated)
The tissue processed via teasing is examined under what?
Phase contrast or bright field: if unstained
differential dyes: if stained
What is the process of squash preparation (crushing)?
Small tissues of tissue (not 1 mm >) placed in 2 microscope slides and compressed forcibly
What type of stain may be placed at the junction of the slide and cover glass w/c is absorbed by capillary action (in squash preparation)?
Vital stain
What is the process (/ principle) of smear preparation?
Examining sections / sediments where cellular materials are spread lightly over a slide by means of a wire loop / applicator stick
What is the purpose of smear preparation?
Useful in cytological exams especially for cancer dx
What are the different types of smear preparation?
1) Streaking
2) Spreading
3) Pull apart
4) Touch preparation (impression smear)
What is the process (/ principle) of streaking?
W/ an applicator stick / wire loop, the material is rapidly and gently applied in a direct / zigzag line
Must obtain a uniform distribution of secretion
What should be avoided when streaking and why should these be avoided?
The formation of too thick / too thin smears because these are unsuitable for exam
What is the process (/ principle) of spreading?
Spread into a moderately thick film by teasing the mucus strands apart w/ an applicator stick
What is the comparison between streaking and spreading?
Spreading is more tedious > streaking
What is the advantage brought by spreading?
It maintains cellular interrelationships
What are recommended sxs where spreading can be done?
1) Fresh sputum
2) Bronchial aspirations
3) Thick mucoid secretions
What is the process (/ principle) of pull apart?
Placing a drop secretion / sediment upon 1 slide over another slide
Slight movement of 2 slides in opposite directions to initiate the flow of materials, 1 single uninterrupted motion
What are the recommended sxs where pull apart can be done?
1) Serous concentration
2) Sputum
3) Enzymatic lavage from GIT
4) Blood smears
What is the process (/ principle) of touch preparation?
Tissue -> in contact and pressed into slide -> cells transferred directly to the slide -> examine via phase contrast microscopy or stained for light microscopic study
What is the advantage of doing touch preparation?
Sx is examined w/out destroying their actual intercellular relationship
