Explain the major limitation of murine monoclonal antibody Flashcards
Murine monoclonal antibody
Antibodies created in rats from identical b cells which are clones from a single parent cell.
First produced in 1975
Limitation
Due to the murine origins of the antibodies they were immunogenic in humans and were poor inducers of immunity in patients
Alternative
Alternative to murine monoclonal antibodies would be chimeric or humanised monoclonal antibodies as their protein sequence are made more similar to human antibodies
Chimeric antibodies
Antibody made up of molecules from domains from different species
Humanised antibody
The conversion of a non-human monoclonal antibody to be as humanlike as possible with respect to immunogenicity, but without losing the antigen specificity and binding affinity of the original antibody
