Enzyme Kinetics Flashcards
What is a catalyst?
Substance that increases the rate of a chemical reaction without being consumed in the reaction
What are the characteristics of a catalyst?
Does not appear in the overall equation
May appear in the rate equation
Does not affect the amount of product formed
What are enzymes?
Protein molecules that catalyse a specific reaction, unchanged by the reactions they catalyse
What is the lock and key model ?
E+S<—> ES—>EP—> E+P
What types of bonds could be present in the enzyme substrate complex?
Ionic
Hydrogen
Van der waals
NOT COVALENT
What does induced fit mean?
Enzyme changes shape
Which step in enzyme kinetics is reversible?
First step
(E+S<—>ES)
Which step in enzyme kinetics is irreversible?
Second step
(ES—>E+P)
What does chemical compatibility mean in enzyme kinetics?
Complimentary sides have to be next to each other to form a complex
What are the assumptions in the Michaelis Menten Model?
Enzyme binds to a single substrate
Two distinct steps
A steady approximation applies
Concentration of substrate is larger
What is Vmax?
Maximum rate that a given enzyme can achieve.
When enzyme active site is activated
What is the turnover number?
The number of substrate molecules converted into product by an enzyme molecule in a unit of time when the enzyme if fully saturated with substrate
What is the equation for turnover number?
K2=Vmax
———
[E]0(s*-1)
What does a bigger turnover number show?
The en payment is quicker at creating the product
What is Km?
Substrate concentration at which the reaction velocity is half of Vmax
