what is cytology used for?
used as a preliminary diagnostic tool
can establish if a lesion is inflammatory and stage the degree, severity, and occasionally etilogy
what can be diagnosed through cytology?
malignancies and hyperplasia
parabasal cell
seen in vaginal cytology
smallest cells
high N/C ratio and basophilic cytoplasm
intermediate cell
seen in vaginal cytology
~2x larger than parabasal cells
superficial/cornified cell
seen in vaginal cytology
small, round nuclei or anuclei, abundant cytoplasm
what types of cells can be seen during proestrus?
a mix: neutrophils, parabasal, intermediate and cornified, mucus
what types of cells can be seen during estrus?
cornified epithelial cells, bacteria
what makes up the ejaculate volume?
- sperm-free watery secretion
- sperm-rich fraction
- sperm-poor fraction
which objective lens is used to observe sperm wave motion?
40x
which objective lens is used to observe sperm motility?
100x
how can we evaluate sperm concentration?
hemacytometer
how can we evaluate live:dead sperm ratio?
stain and smear
sperm morphology
head with acrosome, neck, tail
centesis
ex: abdominocentesis, thoracocentesis, cystocentesis, arthrocentesis
anesthesia and aseptic prep for peritoneal/pleural fluid
make smears, EDTA and beaker of sample
record gross characteristics and cell types
