CULTURE MEDIA Flashcards
● Solid, liquid or semi solid designed to support the growth of microorganisms.
CULTURE MEDIA
What is the percentage of the solidifying agent added
1.5-3% of agar
is the most commonly used
as a solidifying agent
Agar
Agar melting point
≥ 95°Celsius
Agar Resolidify
< 50°Celsius
LIQUID/BROTH USES
○ Can be used to propagate large numbers of microorganisms in fermentation studies
○ For various biochemical tests.
● Contains specific nutrients required for the growth of particular bacterial pathogens
● Used to enhance the growth of particular pathogen from a mixture of organism by nutrient specificity
ENRICHMENT MEDIA
● Favor the growth of a particular bacterium by inhibiting the growth of
undesired bacteria and allowing the growth of desired bacteria
SELECTIVE MEDIA
usually by adding blood, or eggs.
ENRICHED MEDIA
● Contain nutrients to support growth of most non-fastidious organisms without giving any particular organism a growth advantage.
SUPPORTIVE/NON SELECTIVE MEDIUM
● Used to distinguish them from other bacteria growing on the same agar plate
DIFFERENTIAL MEDIA
TUBE MEDIA
● Weigh
● Dissolve
● Dispense
● Sterilize
PLATED MEDIA
● Weigh
● Dissolve
● Sterilize
● Dispense
● The process of rendering a medium or material free of all forms of life
STERILIZATION
● Exposure to steam at 121°Celsius and 15 lbs of pressure for 15 minutes or longer, depending on the nature of the item.
● Microorganisms, even endospores, will not survive longer than about 12 to 13 minutes.
