What is core practical 6? (1)
determining the water potential of a plant tissue.
what is the first step of this practical? (2)
take a potato and cut 6 cylinders, 4cm long using a cork borer. make sure none of the cylinders have peel attached. blot each cylinder carefully to remove excess liquid and weight it.
what is the second step of this practical? (1)
label 5 large boiling tubes one with distilled water, 0.25, 0.5, 0.75 and 1.0.
what is the third step of this practical? (2)
make up 30cm3 of these concentrations of sucrose solution. place one cylinder in each boiling tube and add 30 cm3 of the appropriate solution to each tube.
what is the fourth step of this practical? (3)
leave all of the boiling tubes and their contents in a cool place for 2 hours. after this time carefully pour out the liquid and blot the cylinders. reweigh the dried cylinders and record the new mass taking away from the original mass.
what is the fifth step of this practical? (2)
calculate the percentage change in mass for each cylinder and plot a graph of percentage change on the vertical side and osmotic potential on the horizontal.
