Chapter 43- Additional Testing Flashcards
Common methods for testing for motility
Hanging drop prep, wet prep, and motility media
Wet prep is examined with a __ objective and look for
high dry; cells moving back and forth
Brownian movement
not true motility- shifting of cells w/ little movement related to other cells
Motility media is used if
organism are nonmotile on microscopic exam
In motility media, if growth is limited to stab line,
nonmotile
Diffuse growth throughout motility media means
motile
Motility media uses two tubes of
sulfide indole motility (SIM)
SIM is not recommended for organisms that produce
hydrogen sulfide bc it blackens
Catalase test is performed on
gram-positive cocci and small gram-positive bacilli
How to determine catalase positive on catalase test
there will be bubbles, no bubbles=negative
Coagulase
an enzyme that coagulates plasma; “clumping factor”
Coagulase test performed on
catalase- positive, gram positive cocci
Two versions of coagulase test
Slide coagulase (rapid screening) and Tube coagulase
Positive result on coagulase test looks like
clot formation
Oxidase activity depends on the presence of
cytochrome C oxidase in bacteria
