Mechanism of leucovorin

A

folate substitute
uses reduced folate transporter to enter cell
normal cells express reduced folate transporter
cancer cells only express folate receptor
leucovorin therefore preferentially delivers “folate” to normal cells, allowing higher therapeutic levels of methotrexate to be reached
