What is sibutramine and how can it be used to combat obesity?
A serotonin and noradrenaline reuptake inhibitor, which lowers appetite and increases thermogenesis.
What are the ADRs of sibutramine?
Hypertension and tachycardia
What is orlistat and how does it work?
A gastric and pancreatic lipase inhibitor which decreases dietary fat uptake.
What are the main ADRs of orlistat?
Flatulence, incontinence and steatorrhoea.
How do statins work?
Inhibit HMG-CoA reductase, reducing hepatic cholesterol production, increasing LDL receptor synthesis and causing more clearance of LDL from plasma.
What are some statin ADRs?
Increased transaminase levels, arthalgia, myopathy, headaches.
Name a short acting statin
Simvastatin or pravastatin
Name two long acting statins
Atorvastatin and rosuvastatin
How do fibrates work?
PPAR-alpha agonist, that lowers blood lipid and triglyceride levels.
Name 2 conditions that contraindicate fibrate use
Hypoalbuminaemia and gall bladder disease.
Name ADRs of fibrates
Impotence, dizziness, blurred vision, headache, rash.
Name three fibrates
Bezafibrate, Ciprofibrate, Gemfibrozil
Name a cholesterol absorption inhibitor
Ezetimibe
How does Ezetimibe work?
Blocks NPC1L1 do reduce cholesterol absorption in the gut.
When would cholesterol absorption inhibitors be used?
To cause even further LDL reduction in a patient already successfully using statins.
