Acid Fast Bacteria
complex layered cell wall containing mycolic acid and waxes
(Acid fast cells have a waxy cell wall; mycolic acid in the cell walls of acid fast bacteria makes these organisms resistant to desiccation (drying) and disinfectants
-Only 2 Genera that are acid fast:
1. Mycobacterium
-diseases include Tuberculosis and Leprosy
2. Nocardia
-Pulmonary Nocardiosis (disease similar to Tuberculosis)

Acid Fast Stain- Ziehl-Neelsen (hot method)
This technique uses heat to force the stain through the resistant cell wall
- Primary stain: Carbol Fuchsin (5 minutes on a hot plate)
- Decolorizer: Acid alcohol
- Counterstain: Methylene Blue (3 minutes)

- Decolorizer: Acid Alcohol

- Examination: Fluorescent microscope 150x-200x examine 30 fields
- Acid Fast: yellow-green fluorescence or reddish orange fluorescence
- Non Acid Fast: No fluorescing organisms
