Acid Fast Bacteria

A

complex layered cell wall containing mycolic acid and waxes
(Acid fast cells have a waxy cell wall; mycolic acid in the cell walls of acid fast bacteria makes these organisms resistant to desiccation (drying) and disinfectants
-Only 2 Genera that are acid fast:
1. Mycobacterium
-diseases include Tuberculosis and Leprosy
2. Nocardia
-Pulmonary Nocardiosis (disease similar to Tuberculosis)

Acid Fast Stain- Ziehl-Neelsen (hot method)

A

This technique uses heat to force the stain through the resistant cell wall

  • Primary stain: Carbol Fuchsin (5 minutes on a hot plate)
  • Decolorizer: Acid alcohol
  • Counterstain: Methylene Blue (3 minutes)
