MoA of Clofazimine
A
- Binding to guanine bases of bacterial DNA –> block template function of DNA –> inhibit bacterial proliferation
- Alteration of membrane structure and its transport
- Disruption of mitochondrial electron transport chain

PK of Clofazimine
A
Absorption: 45-62% after oral administration (presence of food may increase rate and extent of clofazimine absorption)
Distribution: Primarily to fatty tissue and cells of reticuloendothelial system
Metabolism: Partially metabolized
Excretion: Partly found in feces (may represent excretion in bile)

Common side effects of Clofazimine
A
Orange-pink discoloration, Ichthyosis, abdominal pain, nausea, diarrhea
