MoA of Streptomycin
Binds to 16S rRNA and 12S protein on 30S bacterial ribosomes –> inhibit further ribosomal protein synthesis –> structural and functional change –> distortion of recognition site of anticodon and codon interaction –> misreading of message carried by mRNA –> cell death (bactericidal aminoglycoside)
PK of Streptomycin
Absorption: Poorly absorbed in GIT; usually by IM or alternative choice in IV
Distribution: Vd 30-35% body weight
Metabolism: Usually not metabolized
Elimination: Primary route –> Urine (Glomerular filtration)
Common side effects of Streptomycin
Allergic reactions, Ototoxicity & vestibular impairment, nephrotoxicity, and neurotoxicity
Side effect management
Ototoxicity, vertigo, headache, and nystagmus –> stop taking streptomycin and change to ethambutol
