3.2 The electron microscope Flashcards
1
Q
What are the 2 main advantages of an electron microscope?
A
1) electron beam has a short wavelength and the microscope can therefore resolve objects well
2) as electrons are negatively charged the beam can be focused using electromagnets
2
Q
Limitations of the TEM.
A
- whole system in a vacuum, can’t observe living organisms
- complex ‘staining’ process
- specimen must be very thin
3
Q
Limitations of the SEM.
A
- whole system in a vacuum, can’t observe living organisms
- complex ‘staining’ process
4
Q
What does a TEM observe?
A
the internal structures
5
Q
What does an SEM observe?
A
the surface and internal structures
