22 - PHARMACOLOGY OF PAIN AND INFLAMMATION Flashcards
SALICYLATES
THEY ARE A CHEMICAL GROUP OF DRUGS EXTRACTED FROM BARK OF SALIX ALBA
THEY INHIBIT COX IRREVERSIBÑY
ASA IS AN ESTER METABOLIZED TO SALICYLIC ACID, THERE ARE OTHER REDIATIES
FROMULATIONS OF SALICYLATES
- BUFFERED-EFFERVESCENT FORMULATIONS; THEY CONTAIN CITRIC ACID AND SODIUM BICARBONATE THAT ACT AS A BUFFER
- CONTROLLED RELEASE FORMULATIONS
- FORMULATIONS FOR TOPICAL APPLICATION
HIGH DOSES OF SALICYLATES
THEY INHIBIT THE REABSORPTION OF URATE BEHAVING LIKE URICOSURIC
ACTIVITY OF ASA
IT HAS PLATELET ANTIAGGREGATORY ACTIVITY AT DOSES LOWER THAN THE ANALGESIC (COX1 INHIBITION MORE POTENT THAN COX2)
ACTION OF ASPIRIN WHEN IT IS TAKEN ALONE
IT IRREVERSIBLY INHIBITS COX-1 ACTIVIY BY ACETYLATION OF A SERINE RESIDUE IN THE ACTIVE SITE OF THE ENZYME
ACTION OF ASPIRIN WHEN IS TAKEN WITH OTHER NSAIDS
WHEN IBUPROFEN AND NAPROXEN IS TAKEN PRIOR TO ASPIRIN, IT CAN OCCUPY THE ACTIVE SITE OF COX AND PREVENT ASPIRIN FROM ACETYLATING COX SERINE RESIUDES. THEIR EFFECT IS RELATIVELY TRANSIENT, NON-ASPIRIN NSAIDS BIND REVERSIBLY.
