Week 2 Flashcards
purpose of week 2
Bacterial Transformation?
uptake of exogenous naked dna of cells.
this new dna will become HERITABLE PART of cells genetic material
DNA?
- deoxyribonucleic acid
- contains genetic material/instructions
RNA
messenger that carries out DNA instructions for controlling the synthesis of protiens
proteins
Monomer= amino acids
Makeup of amino Acid
- acidic group (carboxyl gorup -COOH)
- Amino group (H2N
- R group (varies and determines its identity
Are Bacteria Prokaryotes or Eukaryotes
Prokaryotes
Bacteria composition
- no nucleus
- dna is organized in one double stranded circulor molecul known as NUCLEOID
bacteria host organism we will study in lab
E choli
E choli
Most studied bacteria,
grows extremely fast
harmless under normal conditions
how did we use bacteria in lab
insert new dna in bacteria so it produces whaterver we want to mass produce
ie.. protiens like insulin
modified foods broccolli
bio remediation
Lab experiment
1) labeled one postiive and negative tube
2) added cacls to neutralize negative charge so plasmid can absorb
3) take ecoli from starter plate use inoculate loop of agar plate
4) then add pblu dna and incubate in ice for 15 min\wait
5) then heat shock for 90 second to get bacteria in plasmid
6) place back in ice for 1-2 mn
7)add liquid liria (food)
8)incubate in room temp for 5-15 min
9)plate contents from tube put onto - and + agar plates and incubate at 37 degrees for 24 hours
How will we know bacteria will uptake
growth/colonies
Predictive results from lab
Positive control (with pBlu plasmid):
Ampicillin + X-Gal LB: Bacteria will survive and turn blue.
Ampicillin LB: Bacteria will survive but will not turn blue.
Negative control (no plasmid):
Ampicillin + X-Gal LB: Bacteria will not survive and will not turn blue.
Ampicillin LB: Bacteria will not survive and will not turn blue.
correct test results from lab
Correct Test Results:
Positive control (with plasmid):
Ampicillin + X-Gal LB → + (blue)
Ampicillin LB → + (not blue)
Negative control (no plasmid):
Ampicillin + X-Gal LB → - (no growth)
Ampicillin LB → - (no growth)
what conclusion could u draw if u do not get the predictive results
- cross contamination
- bacteria did not take up plasmid due to innacurate timing of ice before and after heat shock
- broth was not added
