Lab experiment

A

1) labeled one postiive and negative tube
2) added cacls to neutralize negative charge so plasmid can absorb
3) take ecoli from starter plate use inoculate loop of agar plate
4) then add pblu dna and incubate in ice for 15 min\wait
5) then heat shock for 90 second to get bacteria in plasmid
6) place back in ice for 1-2 mn
7)add liquid liria (food)
8)incubate in room temp for 5-15 min
9)plate contents from tube put onto - and + agar plates and incubate at 37 degrees for 24 hours
