P53
Transcription factor for p21; blocks G1–> S phase
- p53 normally upregulates BAX which disrupts bcl-2 which allows leakage of cytochrome C from mt. and activates apoptosis
- both genes must be knocked out for tumor formation
- most human cancers and Li-Fraumeni syndrome(familial cancers of breast, brain, adrenal cortex, etc)
Rb
Inhibits EF2; blocks G1–>S phase
EF2 is released when RB is hyperphosphorylated by cyclinD/CDK4 complex
-RB mutations result in constitutively free EF2 allowing progression thru the cell cycle and uncontrolled growth of cells
Sporadic mutation= unilateral RB
Germaine mutation= bilateral RB and OSTEOSARCOMA
(RB is on chromosome 13)
