Methimazole, Propylthiouracil (PTU)

What is the common MoA of hyperthyroid drugs? What is unique about PTU?
A
Both block thyroid peroxidase, blocking oxidation and coupling of iodine
PTU also works on 5’-deiodinase, blocking peripheral T4 –> T3

Hepatotoxicity

PTU in the first trimester
