Mutated proteins convert other proteins to misfolded proteins which leads to aggregation due to the same shape.

Prion like protein
How does TAU abnormalities cause neurodegeneration?
Mechanisms are still unclear- but suggestions include: oxidative stress, apoptosis, necrosis or simple space occupying lesion.
