RNAi Flashcards
what is hybridization?
the process of combining two complimentary single DNA or RNA molecules to form double molecule
what does RNA hybridization allow for?
the detection of mRNA molecules
what does the injection of dsRNA promote?
mRNA degradation
what is dicer?
a nuclease enzyme that cleaves dsRNA into small fragments called siRNAS
what happens to the siRNAs?
bound by argonaute and other components of RISC complex
what is RISC?
RNA induced silencing complex - ribonucleic acid and RNA-binding proteins
what does argonaute 2 do?
cleaves sense strand of RNA
what does the RISC-siRNA complex do?
binds and degrades complementary mRNA
what is the purpose of RNA interference?
defense against viruses and jumping genes (especially plans, worms, insects)
what are some cell process that use RNAi?
- destroys viral RNA during infection
- preventing translation of mRNA into protein
- activating RISC complex to degrade mRNA for specific genes in lab
what is induced to silence genes of interest?
double-stranded RNA
what is RNA shRNA?
artificial RNA molecule with hairpin turn that mimics dsRNA molecule
what does RNAi allow for?
removing specific proteins without affecting the genome (curing huntingtons????)
helps identify cell morphology proteins
