Repro Technologies Flashcards
How do you use a hemacytomter to calculate the concentration of sperm? The equation is (sperm # in E) X (multiplication factor for E) X (dilution factor) = sperm/mL.
Count the sperm in five of the squares in major square E, find the average of the 5, and multiply this number by 5. The multiplication factor is 10,000 and the dilution factor is 5ju.
How do you calculate the number of breeding units that can be prepared from an ejaculate?
Multiply the sperm concentrations by the ejaculate volume by the motility of normal sperm by the morphologically normal sperm to get the total normal sperm in the ejaculate. Divide this by the desired number of sperm in each breeding unit to get the total number of breeding units. Always round down.
How can you calculate the amount of semen extender needed to process an ejaculate?
You multiply the number of breeding units by the volume of semen in each unit. Then, you subtract the original ejaculate volume.
