Q

what does ras do?

A

regulates diverse cell signalling cascades: proliferation, differention, apoptosis etc via tyrosine kinases

Q

what is the classic ras pathway?

A

grb2 binds to trk using sh2 domain
grb2 recruits sos1 (gef enzyme)
ras gap changes ras gdp for ras gtp

15
Q

how is MAPK linked to cancer?

A

GRB2 is bound to tyrosine kinase receptors and sos binds to GRB2 which activates ras
but if ras mutated then pathway goes ras->raf->mek->erk->mutations in transcribed dna
uncontrolled growth and division etc

16
Q

what are MAPK scaffolding proteins?

A

help control temporal and spatial aspects of MAPK pathway

17
Q

what is most common scaffolding protein of MAPK pathway?

A

kinase suppressor of ras (KSR)

18
Q

what are two proteins involved in temporal regulation of MAPK signalling?

A

EGF and NGF

bith via ERK pathway
