PTSB and transition state analogs Flashcards
1
Q
serine proteases
A
break down proteins
example is chymotrypsin
2
Q
chymotrypsin
A
uses electrostatic catalysis in oxyanion hole.
Anion with a negative charge on one or more oxygen is put in a hole
3
Q
after ES is formed…
A
Have an amide bond.
This amide bond needs to be cut…
why? serine proteases have specificity pocket that allows to determine where to cut. (aromatic amino acid to the left, where it will fit in the pocket)
4
Q
serine protease bacteria
A
- boronic acid deric (TS analog) can interact with serine protease in bacteria, and when this happens, it inhibits the enzyme because its jamming the TS binding site
- in humans, serine protease has a speicificity pocket for aromatic rings, which is why the boronic acid deriv cannot jam itself in the human chymotrypsin to inhibit the enzyme
- would stop digestion if inhibited
5
Q
catalytic triad
A
serine, his, aspartic acid
