Prostaglandins Flashcards
Eicosanoids
Prostaglandins and their related compounds
potent; wide range of physiologic responses
Procoagulant
platelets make thromboxane that makes platelets aggregate and vasoconstriction
Anticoagulant
vascular endothelial cells make prostacyclin that inhibits platelet aggregation and stims vasodilation
Arachidonic Acid
precursor for most eicosanoids
made from linoleic acid (FA)
stored on phospholipids
released by phospholipase (A2)
Lipoxygenase
enzyme that synthesizes leukotrienes
Cyclooxygenase
enzyme that makes prostacyclin and thromboxanes
Glucocorticoids
steroidal anti-inflammatory drug- especially for asthma
promotes up-regulatioon of phospholipase A2 inhibitor
down-regulate COX-2
Where do steroids act?
gene level
NSAIDs
Aspirin
Inhibit COX step
no prostacyclins and thromboxanes made
Aspirin Mechanism
irreversibly inactivates COX by acetylation
suicide inhibitor
Aspirin effect on Platelets
cannot make new COX
no thromboxane production
Aspirin effect on vascular endothelial cells
can make new COX
make prostacyclin at reduced rate
