Pharm MOA only Flashcards
Bevacizumab
Monoclonal Ab against VEGF
-inhibits angiogenesis
Methotrexate
DHFR inhibitor
- Folic acid analogue
- inhibits pyrimidine synthesis
5-Flourouracil
Inhibits Thymidilate synthase
-inhibits pyrimidine synthesis
Cytarabine
Inhibits DNA polymerase
-pyrimidine analogue (CyTarabine)
Azathioprine/6-MP
Inhibit de novo purine synthesis
- Purine analogues
- Metabolized by XO (increased toxicity with allopurinol use)
Dactinomycin
Intercalates DNA
-Kids ACT out (Wilms tumor, Ewing sarcoma, Rhabdomyosarcoma)
Doxorubicin/Daunorubicin
Generates free radicals, DNA intercalator
- AE of cardiotoxicity
- Dexrazoxane used to prevent cardiotoxicity
Bleomycin
Free radical generation
-Pulmonary fibrosis
Cyclophosphamide/Ifosfamide
Alkylates DNA at N-7 Guanine
-hemorrhagic cystitis => mesna
Nitrosureas (carmustine, lomustine…)
DNA alkylators
- Cross BBB (used on brain tumors)
- CNS toxicity
Busulfan
DNA alkylator
-Pulmonary fibrosis
Vincristine/Vinblastine
Inhibit microtubule formation
- binds beta-tubulin and prevents it’s polymerization into microtubules
- Peripheral neuritis, bone marrow suppression
Paclitaxel
Hyperstabilize microtubules so they can’t be broken down
-mitotic spindle can’t be broken down (stuck in M phase)
Cisplatin/Carboplatin
Cross-links DNA
- Nephrotoxicity, Ototoxicity
- Prevent Nephrotoxicity with amifostine (free radical scavenger) and chloride diuresis
Etoposide/Teniposide
Inhibits Topoisomerase II
