PAG - Investigation into the effect of substrate concentration on enzyme activity Flashcards
1
Q
What does the substrate concentration affect
A
how much product is made
2
Q
Catalase
A
Increases the breakdown of hydrogen peroxide
3
Q
Hydrogen peroxide
A
toxin produced as a by-product of reactions in cells
4
Q
where is catalase found in high concentrations
A
raw potatoes
5
Q
Step 1 - preparation
A
- grind 2cm3 piece of potato cylinder with 5cm3 distilled water to make a smooth paste containing the enzyme
- place 10cm3 hydrogen peroxide into a test tube
6
Q
what happens after we have prepared for the experiment
A
- using forceps dip a filter paper disc into enzyme suspension, tapping off excess
- drop filter paper disc into hydrogen peroxide solution, and measure time taken from hitting surface to float up again
7
Q
Health and safety -
A
- Hydrogen peroxide = corrosive = take care
- Glasswear = may smash = handle with care
- Bags/Chairs = tripping hazard = put away
8
Q
Independent variable
A
concentration of hydrogen peroxide - %
9
Q
Dependent variable
A
time taken for discs to resurface - secs
10
Q
Control variables
A
- size of discs
- make sure potatoes are cut to the same size before crushing
- volume of solution
- volume of water mixed with hydrogen raw potato
11
Q
percentage error calculation -
A
uncertainty value / your measurement x100
