Q

process of NGS

A

1) take genome and fragment
2) machines then separate each fragment into their own area of the machine
3) these fragments are amplified using PCR involving fluorescent labelled nucleotides
4) each nucleotide is labelled with a different fluorescent dye
5) the different coloured nucleotides ae added sequentially
6) until order of the nucleotides is sequenced

Q

why is there a need for deep sequencing

A

multiple reading are need to reduce error so we can be sure sequences we are looking at are true reflections
