Features of a light microscope
Expensive to buy and operate. The radiation used is light. It is small and portable. The magnification maximum is times 2000. The maximum resolution is 200 nm. The images are produced is a natural colour is seen or a stain. There is no distortion to the sample. It does not require a vacuum
Features of a scanning electron microscope
It is expensive to buy and operate. The radiation used is an electron. It is large and needs to be installed. The maximum magnification is times 100,000. The maximum resolution is 3 to 10 nm. The image produced is black and white and is coloured Digitally. The sample preparation is complex and often distorts the image. And there is a vacuum needed
Features of a transmission electron microscope
It is expensive to buy and operate. Radiation used is electron. Size is very large and requires installation. Maximum magnification is times 500,000. Maximum resolution is .5 nm. Type of image produced is it black and white and is coloured digitally. Sample preparation is complex and often distort the image. And a vacuum is needed
Advantages of transmission electron microscope is over scanning electron microscope
A transmission electron microscope allows you to serve detail as small as atoms you to high magnification and resolution
Advantages of scanning electron microscope over transmission electron microscope
Large amounts of sample to be observed at one time
Define magnification
Magnification is how large the image is (under a microscope) compare to the actual size

Resolution is the ability to distinguish between two different destructive points on an image

Eyepiece graticule what is it
And eyepiece graticule is it a device used to measure the size of your specimen under a microscope. E.g. a ruler for the microscope.
How is an eyepiece graticule fitted
Can I please graticule is fitted by sliding onto the eyepiece lens
What is a stage micrometre
A stage micrometre is a glass slide paste onto the stage. It’s an accurate scale usually has 100 divisions This leads to each division being 0.01 mm in length and are used to work out the value of each die version onto the eyepiece graticule at a particular magnification
Stages to calibrate the eyepiece graticule
- Place the IP scared you’re in the eyepiece, and placed the stage micro meter on the stage.
- The line 0 point of the Eyepiece graticule with the zero point of the stage micrometre.
- Scanning along with a scale is exactly aligned again.
- Count how many IPC units are equal to the stage micro meter units e.g. 20 EPu equals 10 stage micrometer units (10×10 = 100um)
- From what you’ve calculated in step four you can work out 1EQU division
