Microscopy Flashcards
resolution
ability of a microscope to distinguish between two objects as being separate
what does higher magnification do
increases resolution
what does higher contrast do
gives more detail but doesn’t increase the resolution
light microscopy: reflected light
lighting from above creates the ability to see bigger sizes of cells
Light microscopy: transmitted light
lighting from underneath
contrast enhancing methods of transmitted light in light microscopy
darkfield, phase contrast, differential interference contrast
light microscopy: florescence
exciting of electron to emit a photon of longer wavelengths
confocal flourescence
increases contrast by removing light
electron microscopy benefits and drawbacks
gives higher resolution
no colors, specimens must be dead
electron microscopy: transmission
appears in black and white, lighter where the electrons passed through to hit the camera
electron microscopy: scanning
no sectioning needed but coat the specimen in a heavy metal, often used at lower magnifications
Darkfield
uses a light sample at an angle
Phase contrast
gives higher detailed images from phase shifts in lighting
Differential interference contrast (DIC)
gives a pseudo 3D appearance
