Tricyclic Antidepressant Examples
Amitriptyline, Lofepramine
How do Tricyclic Antidepressant work?
Inhibit neuronal reuptake of serotonin AND noradrenaline (also block a wide range of receptors = side effects)

Second-line treatment of Depression, Neuropathic pain
Tricyclic Antidepressant Contra-indications
Elderly, CVD, Epilepsy, Constipation, BPH, High intraocular pressure
Tricyclic Antidepressant Side Effects
Blurred vision, Dry mouth, Hypotension, Arrhythmias, Convulsions, Hallucinations, Tremor (many)
What drug class shouldn’t you give with TCAs and SSRIs?
Monoamine Oxidase Inhibitors
Selective Serotonin Reuptake Inhibitor Examples
Citalopram, Fluoxetine, Sertraline
Why do SSRIs have fewer side effects than TCAs?
Do not inhibit noradrenaline uptake or cause blockade of other receptors

First-line for depression, Panic disorder, OCD
Who are SSRIs not that good in?
Young people
SSRI Side Effects
Hyponatraemia, Confusion, Suicidal thoughts, Prolonged QT interval, Bleeding
What should you prescribe with SSRIs if also taking Aspirin/NSAIDS?
Gastroprotection due to an increased risk of bleeding
Benzodiazepine Examples
Diazepam, Lorazepam, Midazolam
How do Benzodiazepines Work?
Target GABAA receptor and upregulate it - increases chloride flow into cell and thus more resistance to depolarisation (depresses synaptic transmission)
Benzodiazepine Indications
Seizures, Alcohol withdrawal, Sedation, Severe anxiety/Insomnia
