Major Histocompatibility Complex: Structure and Function- Dr. hudson Flashcards
Can you transfer the product of B cells (serum antibodies) to a non infected (naive) mouse to protect it from a virus?
Yes, it is called passive immunization. Gamma-globulin (IgG) injection for HIV patients, HBsIgs for hepatitis patients. Expensive because antibodies have half life of 30 days so need to harvest often
If you transfer t cells from infected mouse to syngeneic (identical) mouse, they will be protected. If you transfer them to a healthy allogeneic (different strain) mouse, they will not be. Why?
Because t cells always recognize foreign antigen in the context of self.
HLA-A, -B, and -C are the heavy chains of _ molecules
class 1
Six classical gene products.. heterodimers of alpha and beta chains of HLA-DR, -DQ, and -DP are gene products of _ molecules
class 2 MHC
How do class 1 MHC molecules allow for diverse recognition?
Think HLA-A, B, C
They are polygenic- There is HLA-A, B- and C.
They are codominant and polymorphic, which means there is one HLA-A from your mom and one from your dad.
So your genotype can be…
HLA-A2, HLA-A9, HLA-B27, HLA B330, HLA C4, HLAC23 for example
Three classical class 1 MHC gene products
HLA-A, HLA-B, HLA-C
In a class 1 MHC structure, the _ and _ domains of the molecule are what contain the polymorphisms in the population
alpha-1 and alpha-2 peptide binding
In class 2 mhc molecules, _ (number) chains span the membrane,
in class 1 mhc molecules, _ chain spans the membrane
two, one
Which MHC class 2 gene product has the most polymorphisms in the population?
alpha or beta chain?
The beta chain
In class 2 molecules, both chains span the membrane, but the _ and _ domains of the molecule are the peptide binding domains
A1 and B1
What are the four domains of the MHC class 2 molecule?
A1- protein binding B1- protein binding
A2- membrane spanning, B2, membrane spanning. these two contain immunoglobulin fold
Polymorphisms in Class 1 MHC proteins occur in the _ and _ domains
a1 and a2 peptide binding domains
Hepatitis b non responders have which two haplotypes?
HLA-DR7 and HLA-DR3
Homozygotes for these two haplotypes are found almost exclusively in the non responder (NR) population
It’s a problem w/ t cell response not with antigen presenting cell
Ankylosing spondylitis, inflammatory possibly autoimmune disease of verterbral joints. Tissue type with _ haplotype is seen in 93% of individuals with AS
HLA B27
