What do Polyenes do?
They form annular channels, which punch a hole in the membrane and kill the fungus
What are the two Polyenes?
Nystatin and Amphotericin B
What do Polyenes bind to?
Ergosterol
What is use of Amphotericin B limited to?
Life threatening fungal infections
How is Amphotericin B administered?
Intravenously
What is the most serious toxic effect of Amphotericin B?
Renal Dysfunction
What do Azoles inhibit?
The enzyme Demethylase
What does Demtheylase do?
Convert Lanosterol to Ergosterol
What do Azoles do?
The conversion of Lanosterol to Ergosterol means the fungal membrane is defective.
What is Fluconazole primarily active against?
Yeasts
Azole side effects?
Varying degrees of liver toxicity. Risk of cardiac arythmias
How do Allylamines function?
They inhibit an enzyme in the early stages of Ergosterol synthesis.
What Allylamine treats Ringworm (dermatophyte) infections?
Terbinafine
What is Flucytosine?
A potent inhibitor of RNA transcription, and DNA Synthesis
What is Flucytosine and Amphotericin B combined to treat?
Cryptococcal meningitis
